Well if you want to get technical -
1) Huey Dewey and Louie's first appearance in the comics/toons way back had them live with Donald with a letter from "Dumbella" Duck in the 1930s.
2) For years, she was never really fleshed out/seen/mentioned until Don Rosa wrote "The Life And Times Of Scrooge McDuck" , which chronicled the life of Scrooge McDuck.

In that arc, she was renamed Della (with Dumbella as a nickname), and officially stated to be Donald's twin sister.

3) One (Italian i think) comic came up with the idea that Della was an astronaut and was stuck in space, and thus couldn't come home to the boys. This, for the longest time, was the only time Della had ever been mentioned before or since as an adult/as a mom. There were a few 1 page comics of her as a kid.

And finally, we come to 4) With DT17, her first animated appearance, deciding to take that ONE COMIC and work with it.

But for people who aren't obsessed with comics, this is pretty much the first time Della's ever been mentioned. Hell, when the show began, no one expected her to be mentioned at all. In the first episode, a portrait of her (And Dewey's surprised "Mom?") was THE FIRST MAJOR CLIMAX/CLIFFHANGER of the show and made everyone go WHAT THE FUCK
